In many organisms, the expression of genetic information is RNA-mediated. Moreover, some organisms like RNA viruses use RAN molecules as their genetic material. Therfore, we can understand a lot biological charactertics of a specific organism through RNA sequencing.
To sequence RAN, the targeted RAN sequences are converted into complementary DNA (cDNA) molecules. However, some sequencing platforms, such as Oxford Nanopore, allows the targeted RNA to be sequenced directly without cDNA CONVERSION.
By sequenxing total mRNA molexules from a cell under a given condition, the gene expression pattern can be profiled. With gene expression profiling, it is profiling, it is possible to identify genes that are significantly up- or down-regulated under a given condition.
In the gene expression profiling, total RNA samples are extracted, ribosomal RNA are removed, and the remaining mRNA molecules are xonverted to cDNA molecles. However, in the Nanopore platfoam, mRNA molecules are dirextly sequenced by capturing them with a poly- A tail.
